The film opens with a baby dinoshark swimming away from a broken chunk of Arctic glacier that calved due to global warming. Three years later, the dinoshark is a ferocious predatory adult and kills tourists and locals offshore from Puerto Vallarta, Mexico. The protagonist, Trace, is first to notice the Dinoshark and witnesses his friend get eaten, but has trouble convincing people that a creature of such antiquity is still alive and eating people.
After an African dinosaur ancestor of the crocodile is found, Dr. Campbell uses its DNA to create prototypes at Paula Kennedy's Genetic Research Co. (Gereco) lab. However one must be put down after killing someone, another escapes and the information is kept from sheriff Harper. His daughter, county dog catcher Diane Harper, helps her ex (welding artist Tom Banning) with his kid brother Michael's beloved dog. They bump into Campbell and discover the dinocroc is growing, having escaped down a tunnel. Although Gereco hires Australian crocodile specialist and intrepid hunter, Richard 'Dick' Sydney, the beast proves impervious to bullets, so the body count rises alarming.
An American scientist up to no good (as usual) by creating the half-pterodactyl, half-barracuda: Pteracuda. When the creature inevitably escapes, it's up to Sharktopus to stop him.
Aspiring college cheerleader, Cassie Stratford consumes an experimental drug that grants her beauty and enough athletic ability to make the cheer squad. The drug has an unforeseen side effect — Cassie starts to grow and grow and grow!
